KPV (Lys-Pro-Val, CAS 69547-51-9, MW 342.43 Da) is the C-terminal tripeptide of alpha-melanocyte-stimulating hormone (α-MSH). In preclinical research, KPV has been studied for: inhibition of NF-κB nuclear translocation and downstream pro-inflammatory cytokine production (IL-1β, TNF-α, IL-6, IL-8); direct anti-inflammatory activity in IBD models including colitis, Crohn's disease, and ulcerative colitis; intestinal epithelial barrier restoration; dermal wound healing via keratinocyte migration; and in-vitro antimicrobial activity against pathogens including Candida albicans. KPV and BPC-157 address gut inflammation through entirely different mechanisms. KPV works intracellularly via melanocortin receptor-mediated NF-κB suppression — it directly reduces the signalling cascade that produces inflammatory cytokines. BPC-157 works extracellularly via the nitric oxide system, VEGFR2, and FAK-paxillin pathways, with a stronger emphasis on angiogenesis, mucosal protection, and gut-brain axis signalling. Conventional immunosuppressants (steroids, calcineurin inhibitors) broadly suppress immune activity across many cell types and pathways. KPV's proposed mechanism in research models is more selective: it enters cells and directly inhibits NF-κB translocation to the nucleus specifically in inflammatory signalling contexts, without the pattern of global immune suppression seen with steroidal compounds. In cell culture and rodent models, this translates to anti-inflammatory activity at the site of inflammation. Reconstitution: KPV is a small, highly water-soluble tripeptide. Draw 1–2mL of bacteriostatic water into a clean 31G insulin syringe. Inject slowly into the vial and swirl gently until dissolved.
